Ⅰ. 서 론

[구아바의 특징]

구아바(Psidium guajava Linn.)는 옛 잉카인들이 고산지대에서 기르고 즐겨 먹었던 것으로 도금양과(Myrtaceae)의 상록활엽 관목 또는 소교목으로 아메리카 열대지방이 원산이며, 아열대에까지 널리 분포되어 있다. Guava, Guayabo 또는 Kuawa로 불려지며, 과일, 뿌리 및 잎은 오랫동안 민간의약으로 이용되어 왔다.1)

구아바는 관목 또는 교목으로 높이가 3~7m이며 가지를 많이 치고 잎은 마주나며 잎을 누르면 강한 향기가 나는 특성이 있다. 꽃은 지름 3cm정도로 꽃잎이 4개이며 잎겨드랑이에 달리고 대부분 하나씩 피지만 가끔 2~3개의 흰 꽃이 피는 경우가 있다. 열매는 공 모양 또는 달걀을 거꾸로 세운 모양이며 길이 5~12cm, 지름 5~7cm 정도이고 연한 붉은빛으로 익어 향기를 풍기며 작고 단단한 종자가 여러 개 들어 있다. 과육은 즙이 많고 달콤하며 비타민 C가 오렌지 보다 5배 이상 많고 비타민 A, B도 풍부해 과일 중에 “비타민 제왕”이라 불린다. 재배하기 쉽고 기온 및 토양 조건에 대한 적응 범위가 비교적 넓으나 열대에서 아열대의 중간 조건이 생육에 가장 적당한 것으로 알려져 있다.2)

현재 우리나라에서 1991년부터 본격적으로 재배법이 연구되어 제주도, 의령 및 경기도 일대에서 재배되고, 현재 우리나라에 구아바의 토착화는 성공을 하였지만 대부분 관상용으로 사용되거나 열매만을 이용하여 소비되고 있으며, 구아바 잎을 이용한 가공제품은 미미한 수준이다.

구아바의 재배 환경적 특성과 생리적 특성은 지구 온난화로 인한 “기후변화에 대처할 수 있는 매우 적합한 대체작물”이며, 또한 고비용 시설하우스, 가온 등의 추가적인 시설을 전혀 필요로 하지 않는 “무가온 식물”로서 고비용 농산물의 생산 환경을 획기적으로 개선할 수 있는 “저탄소 녹색성장에 적합한 친환경 식물”이라 할 수 있다.

구아바의 열매와 잎은 식용 및 기능성 소재로서 활용가치가 매우 높은 고부가가치 식물로서, 현재 국내에서는 식약청으로부터 구아바 잎 열수추출물이 “식후혈당조절에 도움을 줄 수 있다.”는 건강기능식품으로 개별인정이 되어있고, 일본에서는 구아바차 음료가 혈당조절용 특정보건용 식품으로서 시판되고 있는 등 향후 고소득 작물로서 재배면적이 확대될 것으로 예측된다.3)

의령군에서 특화작물로 육성중인 구아바는 저탄소·친환경·고소득 녹생성장·무가온 대체식물로서 2008년부터 잎과 열매가 생산되고 있고, 매년 생산량이 증가되고 있으나, 이를 활용한 기능성 가공제품의 개발과 상품화사례는 극히 미진한 실정이다.

구아바 열매에는 비타민 A와 C가 사과에 비해 각각 3배와 2.5배 많이 함유되어 있고, 특히 펙틴을 비롯한 식이섬유가 사과에 비해 5.6배, 오렌지보다 약 10배 이상 들어있으며, 또한 칼슘, 철분, 칼륨 등의 무기질과 안토시아닌 색소인 lycopene 등이 풍부하게 함유되어 있는 영양과일이다.3,4,5,6) 구아바 잎은 탄닌을 비롯한 strictinin, isostrictinin등의 폴리페놀이 10%이상 함유되어 있으면서 비타민A, 비타민B1, B2, B6, 나이아신, 바이오틴, 판토텐산, 엽산, 이노시톨 등이 다른 식물소재보다 많이 포함되어있는 기능성 소재이다.5,7,8,9,10,11,12) 구아바 잎의 에탄올 추출물은 생체 내 과산화반응을 저해하는 anthocyan류, alkaloid류, flavonoid류, tannin류와 같은 페놀성 화합물을 함유하며,18,19,20,21,22,23) 그 주요 성분에는 quercetin 배당체인 quercetin 3-O-β-D-glucoside(isoquercitin)

quercetin 3-O-β-D-galactoside(hyperin), quercetin 3-O-β-L-rhamnoside(quercitrin), quercetin 3-O-gentobioside, quercetin 3-O-α-L-ara-binoside(guajavarin)등의 플라보노이드24,25)가 있고 그 외에 β-selinene, β-caryophyllene, squalene, morin-3-O-α-L-lyxopyranoside, morin-3-O-α-L-arabopy-ranoside, β-sitosterol, oleanolic acid 및 ursolic acid26,27,28,29) 등이 함유된 것으로 보고되었다. 옛 잉카인들은 고산지대에 구아바를 재배하여 화분식물은 물론 잎, 나무껍질, 열매 등을 건강식 및 약용으로 이용하였다. 특히 구아바는 탁월한 정장작용으로 설사를 방지하고 위장의 기능을 활성화 시키는 작용이 큰 것으로 보고되어 있다.13) 볼리비아, 이집트, 인도 및 중국에서는 구아바 잎을 감기와 폐결핵 치료 및 항염증과 지혈제로 사용하였으며14,15,16) 멕시코에서는 활성 성분으로 생각되어지는 quercetin과 alycone 화합물들이 설사를 멈추게 한다고 보고하였다.17)

구아바 잎의 주요 생리활성은 여러 연구자들에 의해 보고되었는데,30) 구아바 잎 유래의 주요 생리물질인 strictinin과 isocitrictinin으로 구성된 폴리페놀성 물질이 체내에 존재하는 탄수화물 분해효소(α-amylase, maltase, sucrase)의 활성을 저하시켜 당의 흡수를 억제하는 작용기전등 당뇨병 개선활성에 관한 보고가 많이 있었다.31,32,33,34,35,36)

구아바잎의 탄닌 성분은 세포의 신진대사를 활성화시켜 췌장 기능을 항진함으로써 인슐린의 분비 촉진과 인슐린 저항성을 개선하는 효과가 있음이 밝혀졌다.32) 또한 저용량 streptozotocin으로 유발한 제 1형 당뇨 모델 생쥐에 건조 구아바 잎과 발효 구아바 잎 주정추출물을 투여하면 공복혈당 감소와 내당능 장애개선 효과가 입증 되었으며, 특히 구아바 잎 발효 추출물에서 췌장의 베타 세포 손상을 보호함으로써 혈당 개선 효과가 있음이 보고된 바 있다.36)

구아바 잎에는 항산화 효과가 있는 flavonoid, tannin과 같은 화합물이 보고되었으며,37,38,39,40,41) 그리고 구아바 잎 추출물은 항산화, 항암, 항알러지, 항균효과가 있는 것으로 알려져 있다.42)

일본에서는 구아바 잎의 ellagitannin 등이 암, 심장질환과 같은 바이러스성 만성질환, 동맥경화증 및 혈전증을 예방하는 효과가 있는 것으로 보고되었다.39,42,43) 구아바 잎과 가지의 에탄올, 메탄올, 아세톤 추출물은 식중독 유발, 그람양성균의 생육을 현저히 억제하는 항균활성이 있고,44,45,46,47,48) 특히 구아바 잎과 열매의 열수추출물이 과산화수소로 유도된 신경세포 독성에 항산화 활성과 산화적 스트레스로부터의 신경세포 보호효과를 나타내어 퇴행성 뇌신경질환등의 예방 및 개선을 위한 기능성 소재로서 활용가치가 높다고 보고된 바 있다.49)

구아바 잎 추출물은 전립선암을 억제하고,50) 대장관련 질환의 개선효과, 미백효과 등이 보고되었다.

구아바 잎 발효액은 아토피 피부염 관련 염증반응 유전자 발현과 단백질 생성량은 억제하며 아토피 개선효과가 있음이 밝혀졌다.53) 그리고 구아바 잎 추출물은 탈색과 염색에도 활용가치가 있는 것으로 알려졌다.54,55,56) 구아바 잎의 다양한 기능성 물질과 우수한 생리활성을 이용하여 구아바 잎의 활용성 증대를 위해 가공식품 개발에 응용한 연구가 보고되었다. 먼저 구아바 잎을 이용한 가공식품 개발의 기초자료를 마련하고자 구아바 잎을 발효, 증숙 및 볶음 처리하여 페놀성 화합물의 량과 항산화 활성과의 상관관계, 기호성 등이 보고되었고,57) 또한 구아바 잎 추출물을 크래커 제조시에 첨가하여 항산화 활성이 강화된 크래커 개발,58) 그리고 구아바 잎 분말을 농도별로 첨가한 쿠키59)를 제조하여 품질 특성을 조사한 결과도 보고된 바 있다.

 

 

 

[아토피 피부염 관련 연구현황]

아토피 피부염(atopic dermatitis)은 알레르기 질환의 일종으로 특히 유아기와 아동기에 주로 발생하며 심한 소양감을 동반하는 만성 염증성 피부질환이다. 구미, 유럽에서의 유병률은 20~35%로 매우 높으며 치료와 예방 방법이 개선되고 있음에도 불구하고 계속 증가하고 있다.60) 한국에서도 초등학생까지 아동의 아토피피부염 진단비율이 1995년 13.7%에서 2005년 29%로 두배 이상의 큰 폭으로 증가하였고 중증 환자의 비율도 증가하는 추세에 있는 것으로 보고되고 있다.61)

아토피(atopy)는 그리스어인 a-topos가 어원으로 ‘특이한’, ‘부적당한’ 또는 ‘비정상적인 반응’등의 뜻을 가지고 있으며, 알레르기 반응을 일으키게 하는 면역물질(IgE, immunoglobulin E)을 쉽게 형성함으로써 천식, 알레르기성 비염, 아토피성 피부염 등을 잘 일으키는 유전적 경향을 나타낸다.62) 아토피피부염의 원인이나 발생기전은 아직 확실히 알려져 있지 않으나 유전적, 환경적 요인, 여러 가지 면역학적 이상 등이 복잡하게 연관되어 작용한다고 알려져 있다. 아토피 피부염의 임상적 특징은 크게 소양증(국한성 또는 전신성 가려움), 발진, 만성적 재발 등을 들 수 있다. 혈청 내 IgE 수치 향상, eosinophilia, basophilia의 자발적인 히스타민 분비 증가, CD23 발현 증가, CD8 suppressor 감소, Th2에 의한 IL-4, IL-5 분비 증가 등의 현상을 동반하기도 한다.63) 아토피 피부염은 연령대에 따라 유아형(생후 2~24개월), 소아형(3~4세에서 10세 전후의 소아), 사춘기 및 성인형(만 12세 이후)으로 분류되며 아토피 피부염은 대부분 IgE와 연관된 면역기전에 의해 발병되는데 특정 알러젠에 대한 즉시형 면역반응보다는 T 세포 이상에 의한 지연형 면역 반응이 관여하는 것으로 보고되어있다.64)

Allergy란 외부물질인 allergen과의 접촉에 의하여 과민반응을 나타내는 선천적 또는 후천적 면역현상으로서 천식, 알레르기성 비염, 아토피 피부염 등이 대표적인 allergy 질환들이다.65) 피부는 외부환경으로부터 경계 역할을 하면서 수많은 항원에 접촉하게 되는데 아토피 피부염은 외부 항원과의 접촉에 의해 과민하게 반응하는 면역이상반응으로서 가려움증과 홍반, 각질형성, 수분손실 등을 나타내는 것이 특징인 재발성이 있는 만성 피부질환이다.66) 주로 영유아기에서 흔히 발생하지만 성인까지도 지속되거나 성인이 되어서 새롭게 발병할 수도 있다.67)

일반적으로 알레르기 반응은 생체가 동일한 항원에 반복적으로 접촉하게 되었을 때 항원에 대해 처음에는 발생하지 않았던 과민반응이 나타나는 것으로 이에는 제 Ⅰ형에서 제 Ⅳ형까지 4가지 유형이 존재하는데,68) 이 중 아토피피부염은 대부분 anaphylaxis type 또는 즉시과민반응이라고 불리는 제 Ⅰ형 과민반응에 속하며 IgE와 밀접한 관련이 있는 것으로 보고되었다. 아토피 피부염 환자의 혈중에는 IgE와 호산구의 증가가 나타나는데, 이는 Th2 세포의 활성화와 이에 따른 IL(interleukin)-4, IL-5, IL-13, 등 cytokine의 분비증가와 관련이 있으며, 급성기에서 만성기로 진행될수록 Th1 세포와 여기에서 분비하는 IFN-ɤ가 주도적인 역할을 담당하는 것으로 알려져 있다. 또한 아토피 피부염의 피부 병변에서는 비만세포가 관찰되고, IgE와 결합한 비만세포가 항원과 결합하게 되면 탈과립 과정을 거치며 histamine 등의 매개물질이나 여러 종류의 cytokine들을 방출하여 염증반응을 촉진시킨다.69) 또한 아토피 피부염은 천식이나 알레르기 비염 등의 다른 알레르기 질환으로 발전할 가능성이 매우 높은 것으로 나타나고 있으므로 아토피 피부염의 초기 치료는 다른 알레르기 질환의 예방에도 중요하다고 할 수 있다.70) 아토피 피부염에는 T림프구(T lympnocyte), 항원에 특이적인 Immunoglobulin E(IgE)를 발현하는 수지상세포(dendritic cell), Th1 세포와 Th2 세포에서 발현되는 cytokines 등의 여러 종류의 세포들과 인자들이 관여하게 된다.71) 먼저, 항원과 특이적으로 결합한 IgE는 랑게르한스세포(Langerhans cells)로부터 항원을 T림프구에 전달하는 것을 촉진시키고, 항원을 전달받은 T림프구는 세포매개 면역반응을 유도함으로써 병변을 유발시킬 수 있을 뿐만 아니라, 면역반응을 지속적으로 유지시키는데 중요한 역할을 하게 된다. T림프구는 생성하는 cytokine의 종류에 따라 Th1 세포와 Th2 세포로 구분할 수 있는데, Th1 세포는 IL-2, IFN-ɤ, TNF 등을 분비하여 대식세포를 활성화시킴으로써 지연형 면역반응을 유도하고, Th2 세포는 IL-4, IL-5, IL-6, IL-10, IL-13 등을 분비하여 IgE 생성을 더욱 증가시키고 비만세포(mast cell)와 호산구(eosinophil) 분화를 유도시킴으로써 과민반응을 유발한다.72) 정상적인 상태에서는 Th1 세포와 Th2 세포간의 상호작용으로 균형을 이루면서 면역반응을 유지하지만, 아토피 피부염에서는 T림프구의 Th2 세포로의 전환이 촉진되어 증가된 Th2가 많은 양의 염증성 cytokine을 분비시키고 혈중 IgE의 상승을 가져옴으로써 염증반응을 더욱 촉진시킨다. 게다가 Th2 세포의 반응을 억제하는 역할을 하는 IFN-ɤ를 생산하는 능력은 저하되고 Th1 세포의 증식이 더욱 억제되어 결국 세포매개성 면역의 저하를 가져오게 된다.67) 결과적으로 아토피 피부염의 병변에서는 Th2 세포에 의한 면역 반응이 우세하게 나타나게 되는데, Th2 세포와 Tc 세포는 granzyme B를 발현하여 피부의 염증반응에 관여하게 된다.73) 급성 아토피 피부염에는 Th2가 관여하고, 만성 아토피 피부염에는 Th1이 관여한다. 초기에는 Th2에서 분비하는 IL-4, IL-5가 중요하게 작용하며, 가려움증이 생겨 긁기 시작하면 Th1 세포로 이동하게 된다.74) 아토피 피부염증으로 인한 흉선세포, 내피세포, 기관지 내포세포에서 생산되는 TARC(Thymus and activation regulated chemokine)/CCL17과 RANTES(Regulated upon Activation, Normal T-cell Expressed, and Secreted)/CCL5는 CC chemo kines에 속하며 인체 T세포에 화학주성을 일으키고 기억 T세포의 세포 간 유착에 인테그린 의존성 유착을 유도한다.75)

 

 

면역반응은 감염성 질환으로부터의 인체를 보호하는 기작으로, 외부 물질과 자신을 구별하고 외부 물질을 중화 또는 제거하여 생체의 항상성을 유지시킴. 면역에 관여하는 장기, 세포 및 분자들이 면역계를 구성하며 이들이 서로 협동하고 집합적으로 작용하여 면역반응을 일으킨다.

인체의 면역계는 알러젠, 여러 가지 화학물질 또는 약물 등에 의해 생체의 다른 기관에 독성작용을 나타내지 않는 용량에서도 임파조직과 상호작용을 하여 면역계의 섬세한 균형에 변화를 일으킬 수 있으며 생명유지를 위해 필수적인 이러한 면역반응의 이상 증가 작용으로 인해 생체의 면역반응이 과도하게 진행되면 알러지, 아토피 피부염, 천식 등 여러 가지 염증 반응 등이 초래될 수 있다.77) 외부 물질에 의하여 활성화된 대식세포 (macrophages)는 반응성이 강한 O2-, H2O2, OH., nitric oxide (NO), eicosanoids 및 inflammatory cytokines을 비롯한 여러 물질을 분비함으로서 염증 반응을 촉진시켜 숙주방어기전에 중요한 역할을 하고 있으며, 대식세포의 활성화에 의해 미생물, 감염체 및 종양세포의 성장을 억제한다.78,79) 자유-라디칼 중 NO는 대표적으로 대식세포의 inducible nitric oxide synthase (iNOS) 유전자에 의해 합성되며 면역반응, 혈관확장, 신경전달, 혈소판 응집억제, 세포외 matrix생성의 억제 등 다양한 기능을 매개한다.80) 구아바의 in vitro 염증 억제 효능 평가를 위하여 염증 반응 지표인 NO 생성량을 조사하였다.

면역질환은 알러지 유발물질 (allergen)이 비만세포 (mast cell)나 호염기구 (basophil)의 세포막에 존재하는 Fc receptor에 매개되어 세포의 핵에서 phospholipaseA2나 lipoxigenase 등을 방출시키고 연쇄적으로 arachidonic acid를 생산하여 결과적으로 leukotriene의 세포외 방출, lysosome을 통하여 histamine이 외부로 방출되어 2차적인 면역이상 반응을 유도하거나 면역 관련의 세포에 손상을 일으킨다.81,82) 과민성면역질환인 알러지 반응은 IgE 항체로의 class switch를 유도하는 cytokine인 IL-4 및 eosinophil의 증식 및 분화를 촉진시켜 eosinophilia를 초래하는 cytokine인 IL-5가 필수적으로 관여하여 근원적 원인인 IgE 항체를 생성한다.83)

 

 

 

 

알러지성 질환은 IgE에 의해 매개되고 Type 2 T-helper (Th2) cell, 비만세포 및 산성백혈구 (eosinophil)가 알러지 유발 과정에 중요한 역할을 하며 알러젠에 의해 활성화된 Stat6-매개신호경로를 통한 IL-4의 생성을 통해 Th2 세포들은 IL-4 및 IL-5 등의 알러지와 관련된 cytokine을 분비하게 된다. IL-4 및 IL-5는 B세포에 의한 IgE 및 IgGI의 생성을 유도하고, 골수에서의 산성백혈구의 발달을 자극하여, 염증이 생긴 조직으로 모이도록 유도하고 Th2 세포가 관여하는 Th2 면역반응에 대한 과민성 면역질환의 유도에 핵심적인 역할을 한다.

in vitro 항알러지 활성을 조사하기 위하여 비만세포에 시료에 의한 IgE의 생성량 변화에 대한 영향을 측정하고, 알러지 반응과 관련된 cytokine인 IL-4와 IL-5의 생성량 및 유전자 발현을 조사하고자 한다. 현재까지 국내․외적으로 인체 아토피성 피부염과 동일한 질환 모델 동물은 없으며, 가장 유사한 모델로 많은 연구자들이 아토피성 피부염 연구에 NC/Nga mouse를 가장 보편적으로 사용하고 있다. NC/Nga mouse는 자발적 아토피성 피부염 유발 모델로 잘 알려져 있으며, 그 원인으로는 JAK3 kinase의 과활성으로 인한 IgE의 생성이 항진되어 나타난 결과로 보고 된 바 있으나 현재까지도 정확한 기전은 알려져 있지 않으며 이런 기전상의 불분명성에도 불구하고 인간의 아토피성 피부염을 가장 잘 반영하는 실험모델로 약리실험에 널리 이용되고 있다.86) 일본에서는 hapten 형성 물질인 1-chloro 2,4,6-trinitro benzene (picryl chloride)을 이용하여 피부염을 인위적으로 유도하여 이러한 문제를 해결하고 있으나, picryl chloride의 경우 국내 반입이 금지되어 있으므로, 구조적으로 picryl chloride와 유사하여 같은 기전으로 작용하는 1-chloro 2,4-dinitrobenzene (DNCB)을 이용하여 시험을 진행하였다. DNCB를 이용하여 아토피성 피부염의 주된 증상들 (가려움, IgE증가, 피부염증)을 관찰할 수 있고, 아토피성 피부염의 치료제에 의해 증상이 감소되는 현상을 관찰할 수 있다.84,85)

피부에서의 염증반응 과정은 Th2 chemokine들에 의존적으로 일어나며, chemokine 들은 면역세포들을 조절하는데 관여함. TARC는 Th2 유형의 chemokine으로 피부세포, 수지상세포, 내피세포, 상피세포 등에서 만들어진다. Macrophage-derived chemokine (MDC/CCL22)은 TARC와 밀접하게 관련되어 있으며, 수지상세포, B 림프구, 대식세포, 피부세포, 내피세포 등에 의해 만들어지는데, 최근의 연구들에 따르면 아토피 피부염 환자의 혈청내 TARC와 MDC chemokine의 양에 따라 아토피 피부염 유발이 결정된다고 알려져 있으며, 이들 chemokine들에 의해 여러 가지 질병들이 조절된다. Cutaneous T cell-attracting chemokine(CTACK/CC

27)는 피부에서 선택적으로 발현되며 아토피 피부염을 조절하는 chemokine인 것으로 보고되었다.88)

본 실험에서는 구아바 발효 에탄올 분획에 대한 아토피 피부염의 억제 효능을 평가하였다. 현재 아토피 피부염 치료에는 보습제의 사용과 함께 국소스테로이드(corticosteroids) 제제가 가장 널리 사용되고 있는데, 단기간 사용하면 매우 효과적이지만 장기간 사용하게 되면 교원섬유의 위축으로 피부가 얇아지며 투여를 중단하면 증상이 악화된다는 부작용이 보고되었다.69) 또 다른 약물로는 면역억제제인 tacrolimus(상품명 Prograf)와 pimecrolimus(상품명 Elidel)가 사용되고 있으나 역지 장기간 사용하면 면역억제 효과에 의한 피부암의 발생 가능성이 있다고 알려져 있다.70)

현재 아토피 피부염의 증상 완화를 위하여 사용되고 있는 치료방법으로는 환경개선, 스테로이드 외용제 처방, 보습제, 항히스타민제, 항생제 등이 사용되고 있으나 항히스타민제를 장기간 사용할 경우 내성이나 저혈압, 피부염, 소화기장애 등의 부작용을 일으킬 수 있어 지속적인 사용은 어려우며, 특히 스테로이드 외용제를 장기간 복용할 경우에는 고혈압, 심부전, 피부위축 또는 소아 환자의 성장 지연 등의 심각한 부작용이 일어날 수 있으므로 사용이 더욱 제한된다.89) 이처럼 발병환자의 삶의 질을 떨어뜨리며 근본적인 치료방법이 없는 아토피 피부염의 증상 완화를 위하여 부작용을 최소화 하고 장기간 사용 할 수 있는 천연물 자원을 탐색하고 개발하는 것이 시급한 것으로 판단된다.

본 연구에서는 구아바 잎과 열매의 일반성분과 항산화물질을 조사하고, 구아바 잎 유래 추출물을 이용하여 in vitroin vivo 아토피 피부염 개선활성, 알러지 억제활성을 검증한 결과를 기초로 구아바 발효액 제품의 항아토피 활성을 규명하였다. 그리고 구아바 발효액을 이용하여 과립차를 개발하여, 그 특성을 조사하였다.

 

 

 

 

Ⅱ. 재료 및 방법

 

1. 실험재료

 

구아바 잎과 열매, 그리고 구아바 발효액 등은 경남 의령군 소재 구아바 코리아(주)에서 제공받아 사용하였다. 구아바 잎은 수분함량 10% 이하로 건조하였고, 구아바 열매는 동결건조 하였으며, 구아바 발효액은 구아바 잎과 줄기에 설탕을 동량 가하여 혼합 후 실온에서 2년간 숙성한 발효액을 사용하였다. 구아바 발효액의 활성 측정용 시료는 동결건조하였다. 그 외에 실험에 사용한 모든 시약은 특급을 사용하였다.

 

2. 연구방법

 

1)일반성분

 

일반성분은 AOAC법90)과 식품공전의 분석방법에 따라 3회 분석하여 평균값으로 산출하였다. 즉, 수분 함량은 105℃ 상압건조법, 회분 함량은 550℃에서 직접회화법, 조단백질 함량은 micro-kjeldahl법을 이용한 단백질 자동분석기(Kjeltec protein analyzer, Tecator, Sweden)로, 조지방 함량은 Soxhlet법을 이용하여 측정하였다.

 

 

2)무기질 측정

 

무기질(Ca, P, Mg, K, Na, Fe, Zn, Cu, Mn)함량은 AOAC법91)에 준하여 분석하였다. 즉, 시료를 0.1mg 단위까지 정확히 칭량하여 550℃에서 6시간 동안 회화시킨 다음, 20℃ sand bath 상에서 5mL의 HNO3 용액을 가하여 10분 동안 가온하고 방냉 후, 25mL volumetric flask에 넣고 증류수를 가하여 여과하면서(whatman filter paper No.41) 정용하였다. 여과액을 적절한 농도로 희석한 후 Inductively Coupled Spec-trometer(ICP, Lactam 8440, Plasma Lab., Australia)를 이용한 유도결합 Plasma 방출분석법으로 분석하였으며, 분석조건은 approx-imate RF Power가 1,150W이며, analysis pump rate는 100rpm으로 하였고, nebulizer pressure와 observation height는 각각 30psi 및 15mm으로 하였다.

 

3)비타민 분석

 

비타민 함량은 식품공전의 분석방법91)을 응용하여 HPLC로 분석하였다. HPLC로 분석하기 이전에 vitamin C, B1, B2, B6, nicotinic acid를 0.1g씩 각각 100mL 플라스크에 넣어 3차 증류수로 정용한 후, 용해시켜 표준농축용액을 제조하였다. 이것을 3차 증류수로 희석하여 적정 농도의 표준용액을 만들어 0.45ɥm MILLEX-HV13 필터(Millipore)로 여과하여 사용하였다. HPLC는 270nm UV검출기가 부착된 장비(Hewlett Packard 1090 series)로서 column은 shisedo C18(5ɥm, 4.6×250nm, Tokyo, Japan)을 사용하였다. 이동상은 A상과 B상으로 사용하였는데 A상은 PIC B7(1-heptanesulfonic acid sodium salt) 1.25mM 및 acetic acid 1%를 용해한 수용액이며, B상은 PIC B7 1.25mM 및 acetic acid 1%를 용해한 60% methanol를 사용하였다. 이동상의 유속은 0.5mL/min으로 0~1분 사이에는 100% A 이동상을, 1~25분까지는 순차적으로 100% B이동상이 되도록 흘려주었으며, 25~28분 사이에는 100% B 이동상으로, 끝으로 10분간 100% A 이동상을 흘려주었다. 시료용액의 주입량은 20ɥL이다.

 

4)총 폴리페놀 함량 측정

 

구아바 잎과 열매의 총 폴리페놀 함량 측정은 Folin-Denis방법을 변형하여 실시하였다.92) n-Hexane으로 탈지한 시료 15g에 70% 메탄올 150ml을 넣고 균질화시킨 다음 90℃에서 30분간 환류냉각한 후 여과하고 남은 잔사에 150ml의 메탄올을 넣고 다시 균질화, 환류냉각 및 여과의 과정을 3회 반복하여 얻은 여과액 300ml를 감압농축시켜 150ml로 한 다음 11,000rpm에서 15분간(5℃) 원심분리시켜 얻은 상징액을 총 폴리페놀 함량 측정용 시료로 사용하였다. 얻어진 시료 5ml에 Folintldir(1/3 희석액) 5ml를 가하고 3분 후 10% sodium carbonate 5ml를 넣어 30℃에서 1시간 발색시킨 다음 700nm에서의 흡광도를 측정하였다. 대조구로서는 검액 대신 물을 사용하였고, 미리 gallic acid를 사용하여 구한 검량곡선으로부터 시료 중의 폴리페놀 함량을 측정하였다.

 

5)카로티노이드 함량

 

총 카로티노이드 함량은 건조분말 2g을 50% 메탄올로 추출하여 수용성 색소를 제거한 후 잔사를 상온에서 아세톤으로 2회 추출하였다. 추출액을 감압농축하여 40℃에서 농축하고 디에틸에테르와 포화염화나트륨용액으로 세척하였다. 상등액을 20ml로 농축하고, 상온에서 10% 에탄올성 수산화칼륨 용액으로 24시간 검화시킨 후, 포화염화나트륨용액으로 3시간동안 분리시켰다. 상등액을 무수 황산나트륨으로 탈수시키고 40℃에서 농축하였다. 클로로포름에 용해한 후 465nm서 흡광도를 측정하여 표품 β-carotene을 이용하여 총카로티노이드 함량을 구하였다.93,94)

 

6)in itrov 항아토피 피부염 및 항염증 활성

 

(1)구아바 잎 추출물 조제

in vitroin vivo 아토피 피부염 개선활성에 사용된 구아바 잎의 시료는 다음과 같이 조제하였다.

열수추출물은 시료 대비 증류수 15배를 가한 후 95℃, 4시간 추출, 여과, 감압농축하여 동결건조하였다. 에탄올 및 메탄올 추출액은 시료 대비 5배의 알콜을 가한 후 상온에서 90시간 침지한 후 여과, 감압농축한 다음 동결건조하였다. 유기용매 분획별 구아바 잎 추출물은 극성도에 따라 순차적으로 분획하였다. 즉 구아바 잎 메탄올 농축액 50ml에 증류수를 3배 가한 후 hexane, choloroform, butanol, ethylacetate를 순차적으로 200mlTlr 가한 후 5회 반복 추출한 다음 65℃에서 감압농축하여 건조된 시료를 사용하였다.

 

(2)세포배양

Human keratinocyte cell line HaCaT는 10% FBS, 2mM L-glutamine, 10ɥ/mL penicillin 및 10ɥg/mL streptomycine이 함유된 DMEM 용액에서 37℃, 5% CO2 조절 배양기로 배양하였다.

DMSO로 용해한 PGEA의 stock solution을 TNF-ɑ/IFN-r를 첨가하기 전에, 30분동안 배지에 처리하였다. 대조구는 DMSO만 처리하였다. Rat mast cell line RBL-2H3은 ATCC(Bethesda, MD)에서 구입하여 10% FBS, 2mM L-glutamine, 100ɥ/mL penicillin, 100ɥg/mL streptomycine이 함유된 EMEM에서 37℃, 5% CO2 배양기로 배양하였다.

 

(3)세포독성 분석

PGEA 세포독성은 wsi-1 assay kit를 사용하여 분석하였다.

즉 HaCaT 세포를 96-well plate에 분주한 다음 10% FBS-DMEM용액에서, 37℃, 24시간 배양한 후 0.1~100ɥg/mL의 PGEA를 첨가하였다. TNF-ɑ 및 IFN-ɤ 20mg/mL를 PGEA 처리 유무에 따라 배양하여 세포독성은 550nm에서 흡광도를 측정하여 산출하였다.

(4)chemokine 생성량 측정

PGEA 10~5ɥg/mL 농도로 30분간 전처리한 HaCaT cell에 TNF-ɑ 및 IFN-ɤ 20mg/mL를 24시간 처리하였다. 상등액을 수획하여, TARC은 sandwich immunoassay을 사용하여 정량하였다.

(5)역전사(reverse transcription : RT)․PCR 제조

HaCaT cell를 PGEA 10~50ɥg/mL로 30분간 전처리한 후, TNF-ɑ 및 IFN-ɤ 20mg/mL로 24시간 배양하였다. 총 RNA는 RNAiso reagent(Takara, Japen)로 추출․분리한 후 다음과 같은 prima를 사용하여 PCR를 행하였다

(6)형질전환 및 luciferase 활성 촉진

혈청과 항생물질무첨가 lipofectamine시약을 사용하여 NF-kB inciferase

repartee 함유 PCMV-β-gal plasmid를 형질전환 시킨 다음 4시간 후에

배양배지에 옮겼다.

형질전환된 세포에 PGEA(10~50ɥg/mL)를 30분간 전처리한 후에 TNF-ɑ

/IFN-ɤ 각 20ɥg/mL를 24시간 처리한 다음 lysis하였다.

이렇게 얻어진 세포추출물을 luciferase 및 β-galactosidase 활성측정에

사용하였다.

 

 

(7) western blot 분석

HaCaT cell PGEA(10~50ɥg/mL)로 30분간 전처리하고, TNF-ɑ/IFN-ɤ

(20ɥg/mL)로 15분(nucleus p65 및 phospho-STATI 분석), 30분(phospho

-p65/IkB-ɑ분석), 6시간(nucleus Nrf2), 24시간(HD-1분석)동안 각각 처

리하였다. 세포 단백질용액(80ɥg)으로 10% sns-PACE를 행한 후 PVDF

menbrane에 전이시킨 다음 5% skim milk로 비특이적 항원항체 반응을

차단시켰다. Membrane에 peroxidase-conjugated anti-IgG secondary

antibody를 붙여 반응시킨 후 ECL시약으로 단백질 밴드를 확인하였다.

 

(8)β-Hexosaminidase 방출량 측정

IgE-sensitized RBL-2H3 cell에 PGEA(10~50ɥg/mL)과 DNP-BSA를 처

리한 상등액과 0.1M sodium acetate bugger(pH4.4)에 녹인 1.2mM 4-

methylumbellife-ryl-N-acetyl-β-D-glucosaminide를 37℃, 30분간 반응

시킨 다음 0.1% Triton-X100으로 세포를 파괴시켰다.

상등액 중의 β-hexoaminidase 활성은 spectrophotometer에서 흡광도(40

5nm)를 측정하여 산출하였다.

 

(9)Histamine 생성랑 측정

IgE-sensitized RBL-2H3 cell에 시료를 처리한 상층과 메탄올에 녹인

1% o-phthalaldehyde, 1N NaOH를 상온에서 5분동안 반응시킨 다음

반응을 정지시키기 위해 1N HCI을 넣은 후 excitation 360nm, emission

450nm에서 Histamine의 생성량을 측정하였다.

 

 

 

7)In viov 아토피 피부염 억제 활성

 

(1)실험동물 및 처리

아토피 피부염 in vivo 억제 활성실험에 사용된 동물은 수컷 6주령 NC/Nga mice로서 SLC사(shizuoka, Japan)로부터 구입하였다. purina korea와 물을 자유롭게 먹도록 하였으며, 사육장의 온도는 21~24℃, 상대습도는 40~80%로 유지시켰고, 12시간마다 낮과 밤이 반복되도록 사육장내 빛을 조절하였다.

NC/Nga 마우스를 실험실에서 2주 순화완료 후 양쪽 귀를 제모하여 피부 병변이 없음을 확인 후 투여 및 피부 도말을 개시하였다.

 

(2)아토피 피부염 유도 및 구아바 추출물의 처치방법

6주령 NC/Nga 마우스를 2주간 안정화 단계를 거친 후 1% 또는 0.2%의 DNCB는 등과 귀에 도포하여 아토피 피부염을 유발하였으며, 그 후 구아바에 대한 감소효과를 알아보기 위해 구아바를 농도별로 등과 귀에 도포하였으며, 아토피 피부염의 유도 및 구아바 추출물의 처치방법은 아래와 같고 실험모식도는 Fig.1과 같다.

(3)구아바 화장수 제조

실험동물에 도포한 구아바 화장수 조성물은 구아바 추출물 100, 200mg

%, 폴리에틸렌글리콜(400) 40%, 에탄올 30%, 정제수 잔량으로 구성하였

다.

 

(4)Dermatitis soring

NC/Nga 마우스의 피부염은 아토피성 피부염에서 일반적으로 사용되는 임상적 육안 평가법을 이용하였다. 육안평가 결과는 홍반 (erythema), 출혈 (hemorrhage), 건조피부 (dryness), 흉터(scarring) 찰상 (excoriation), 짖무름 (erosin) 등 6가지 항목을 평가한 점수의 합으로 나타내었다. 각각의 항목은 없음 (0), 약함 (1), 중증도 (2), 심함 (3)으로 채점하였다.

 

(5)마우스 비장의 무게 및 체중변화

매 주 1회 마우스에 처리할 때 체중을 측정하고 비장을 수거하여 칭량하였다.

(6)혈청 IgE 측정

실험동물의 꼬리 정맥과 안와 동맥으로부터 채취한 혈액을 원심분리 한 후 혈청 IgE 농도를 sandwich ELISA 방법을 이용하여 측정하였다. 시료를 96well plate에 50mM carbonate buffer로 희석한(× 1000) 1st 항체를 100μL씩 분주한 후 4℃에서 overnight시겼다. 1/4 block buffer를 300μL씩 분주하여 실온에서 30분 반응시킨 후 항체가 함유되어 있는 시료를 20μL씩 각 well 에 첨가한 후 실온에서 2시간 반응시켰다. 효소 표식한 2nd biotin-conjugate antibody 100μL를 각 well에 첨가한 후 실온에서 2시 간 반응시켰다. HRP-streptavidin conjugate를 100μL씩 넣고 실온에서 30분 배양한 후 기질용액을 각 well에 100μL씩 넣고 37℃에서 20-30분 동안 반응시켰다. 각 단계는 0.05% PBS-Twin (Invitrogen Corporation, Grand Island, NY, USA)으로 세척하였으며 마지막으로 발색 후 1.5% oxalic acid를 100 μL넣고 반응을 정지시키고, ELISA reader(405nm)로 측정하였다.

(7)혈청 IL-4, IL-5, IFN-γ 측정

실험동물의 안와 동맥으로부터 채취한 혈액을 원심분리한 후 혈청 IL-4, IL-5, IFN-γ 농도를 sandiwich ELISA 방법을 이용하여 측정하였다. 시료를 96well plate에 PBS로 180배 희석한 capture antibody를 100μL씩 분주하여 실온에서 overnight 시킨 후, block buffer 300μL을 분주 하여 최소1시간 실온에서 반응시킨 다음 reagent diluent로 희석한 각 샘플과 표준액을 100μL씩 분주한 후, 실온에서 2시간 반응시켰다. Streptavidin-HRP을 working dilution으로 희석하여 100μL씩 각 well에 분주하여 실온에 20분 반응시킨 후, substrate solution을 100μL씩 분주하고 다시 실온에 20분 반응시켰다. 각 단계는 0.05% PBS-Twin으로 세척하였으며 마지막으로 발색 후 2NH2SO4를 50μL씩 넣고 반응을 정지시킨 다음 동일한 과정을 통하여 purified cytokine을 표준으로 하여 ELISA reader(405nm)로 측정하였다.

 

(8)비장 임파구 배양 분리, 배양 및 비장임파구의 IgE 측정

실험동물을 경추탈골로 희생시켜 비장을 적출하고 비장임파구는 RPMI 1640 medium(Invitrogen Corporation) 내에서 cell strainer로 분리하였다. 임파구 세포에 RPMI 1640 medium을 첨가하여 300×g,4℃에서 5분씩 3번 원심분리한다. 임파구를 분리한 후 RPMI 10% FBS(Invitrogen Corporation)가 함유된 RPMI 1640 medium에 부유시켜 24well plate에 2 × 106cells/well의 농도로 분주하고, 임파구의 증식을 촉진시키기 위하여 125μg/mL의 농도로 PBS에 용해시킨 concanavalin A를 10% 첨가한 후 5% CO2, 37℃ incubator에서 48시간 배양하였다. 배양한 임파구 상층액의 IgE 함량을 sandwich ELISA 방법을 이용하여 측정하였다. 총 항체 양의 측정으로는 항체의 F(ab)2단편을, 항원 특이적 정량법으로는 항원을 ELISA plate에 고정한다음 배양상층(supernatant)을 반응시켰다. 항체는 ELISA plate 표면에 비특이적으로 결합하는 경향이 있기 때문 에, 고정항체 또는 항원을 고정시킨 후 단백질용액으로 처리하는 blocking 조작을 행한 다음 고정항체 또는 항원에 결합한 항체에 효소 표식한 항체를 결합시켜, 기질용액을 첨가하여 발색시켰다.

IgE는 검출감도를 높일 필요가 있기 때문에 avidin-biotin법을 사용해 ELISA법으로 검출하였다.

 

(9)조직병리학적 검사

마우스의 양쪽 귀 피부조직은 생검하여 10% 포름알데히드 용액에 고정하여 조직 염색 전까지 4℃에서 보관하였다. 피부 조직은 피부의 전반적인 상태, 비만세포의 침윤과 탈과립화, 호산구의 침윤을 중심으로 관찰하였다. Hematoxylin-eosin 염색법을 이용하여 조직을 염색하고, 광학현미경상에서 약 200배 배율로 관찰하였다.

 

 

(10)대식세포 활성화에 대한 영향

실험동물에 시료를 투여한 다음, 복강 대식세포를 분리하여 대식세포의 수를 비교한 뒤 대식세포의 활성화를 확인하였다. 대식세포 탐식능 (식세포 작용; phagocytosis) FITC-zymosan method를 측정하였고 Nitric Oxide 측정은 실험동물에 시료를 투여한 다음, 복강 대식세포를 분리하여Griess시약을 이용하여 NO를 측정하였다.

 

8)통계처리

 

모든 실험 결과들은 mean±S.E.M.로 나타냈고, 통계처리는 Student's t-test를 실시하여 p〈 0.05를 기준으로 유의성 여부를 판정하였다.

 

 

 

 

 

 

 

 

 

 

 

 

 

Ⅲ. 결과 및 고찰

 

1.구아바 잎 및 열매의 일반성분

 

본 실험에 사용된 건조 구아바 잎 100g과 구아바 생 과일 100g에 대한 일반성분을 조사한 결과는 Table 1과 같다.

건조 구아바 잎의 일반성분은 수분 7.01%, 탄수화물 15.65%, 조단백질 5.48%, 조지방 2.16%, 회분 9.12%로 구성되어 있었는데, 탄수화물과 회분 함량이 높은 특징을 보였고 조단백질과 조지방도 다른 식물체 잎으로부터 높은 영양특성을 나타내었다.

구아바 열매의 일반성분의 특징은 탄수화물이 17.41%로서 가장 높았고, 조단백질, 회분, 조지방 순으로 검출되었다. 그리고 구아바 잎과 열매의 식이섬유 함량은 각각 30.28%와 3.45%로서 잎에서 높은 함량을 보였다.

2. 구아바 잎 및 열매의 무기질 함량

 

건조 구아바 잎과 구아바 생과일의 무기질 함량은 Table 2와 같다. 과일보다 잎에서 무기질 함량이 전반적으로 높았고, 잎에서는 Ca이 1925.23mg%로서 매우 높았고 K, Na, Mg, P, Fe 순으로 검출되었다. 과일에서는 K가 314.62mg%로서 가장 높게 검출되었고, Na, P, Ca, Mg, Fetns으로 높게 나타났다. 구아바 잎은 칼슘의 급원으로서 활용가치가 높고, Mg, Fe등도 타 식품에 비해 높은 특징을 보여 우수한 식품소재임이 밝혀졌다.

3. 구아바 잎 및 열매의 비타민 함량

 

건조 구아바 잎과 구아바 생과일의 비타민 함량은 Table 3와 같다. 구아바 잎과 열매 모두에서 비타민C가 각각 13.27mg%, 45.28mg%로서 높게 나타났고 niacnin이 각각 32.55%, 41.17%로서 가장 높게 함유되어 있었다. 그 외에 비타민 B1, B2, B6도 골고루 함유되어 있는 것으로 나타나 비타민 급원으로서도 우수한 소재인것으로 사료된다. 국가별 구아바 열매의 비타민C 함량은 미국이 240mg%, 일본이 120~300mg% 로서 의령군 구아바보다 높은 것으로 보고되었다.

4. 구아바 잎 및 열매의 항산화 물질

 

건조 구아바 잎과 구아바 생열매로부터 주요 항산화 물질로 알려진 폴리페놀성 물질과 카로틴노이드 함량을 측정하였다(Table 4). 건조 구아바 잎은 생열매에 비해 항산화성 물질인 폴리페놀과 카로틴노이드 함량이 전반적으로 낮았고, 잎에서는 카로틴노이드 물질이 거의 검출되지 않았다. 구아바 생열매에서는 총 폴리페놀 함량이 490mg%, 총 카로틴노이드가 215μg%로서 항산화성 물질의 함량이 높은 특징을 보였다.

5. 구아바 잎의 in vitro 아토피 피부염 억제활성

 

구아바 잎으로부터 추출한 열수추출물(GW), 메탄올 추출물(GM) 그리고 ethylacetate 분획물(GEA)을 건조하여 in vitro 아토피 피부염 억제활성 즉, TARC/CCL 17의 발현능, in vitro 염증억제활성, in vitro 알러지 억제활성 정도를 조사하였다.

 

1)TARC/CCL 17의 생성능과 유전자 발현능

 

인체 각질 세포에서 여러 가지 사이토카인 중 TNF-α와 IFN-γ에 의해 TARC/CCL17에 의해 생성이 증가하므로 인체 피부 각질 세포인 HaCaT 세포에 TNF-α를 처리하여 아토피 지표인 TARC/CCL17을 유도시킨 후 구아바 잎 열수추출물(GW), 구아바 잎 메탄올 추출물(GM) 및 구아바 잎 에틸아세테이트 추출물(GEA)에 의한 TARC/CCL17의 생성에 대한 영향을 측정하였다. TNF-α에 의해 증가된 TARC의 유전자 발현과 단백질 생성량이 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50 ug/ml)처리에 따라 농도 의존적으로 감소하였다(Fig. 2, 3).

또한 HacaT 세포에 TNF-α를 처리하여 구아바 열수 추출물 및 메탄올과 에틸아세테이트의 RANTES/CCL5 생성량에 대한 영향을 조사한 결과 TNF-α에 의해 증가된 RANTES의 생성량이 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)처리에 따라 농도 의존적으로 감소하였다(Fig. 4).

2)In vitro 염증 억제활성

 

(1)Nitric oxide 생성능 조사

면역반응은 감염성 질환으로부터의 인체를 보호하는 기작으로, 외부 물질과 자신을 구별하고 외부물질을 중화 또는 제거하여 생체의 항상성을 유지시킨다. 면역에 관여하는 장기, 세포 및 분자들이 면역계를 구성하며 이들이 서로 협동하고 집합적으로 작용하여 면역반응을 일으키는 것으로 알려져 있다(Littman and Pamer, 2011).

인체의 면역계는 알러젠, 여러 가지 화학물질 또는 약물 등에 의해 생체의 다른 기관에 독성작용을 나타내지 않는 용량에서도 임파조직과 상호작용을 하여 면역계의 섬세한 균형에 변화를 일으킬 수 있다. 생명유지를 위해 필수적인 이러한 면역반응의 이상 증가 작용으로 인해 생체의 면역반응이 과도하게 진행되면 알러지, 아토피 피부염, 천식 등 여러 가지 염증 반응 등이 초래될 수 있다(Jutel and Akdis, 2011). 외부 물질에 의하여 활성화된 대식세포 (macrophages)는 반응성이 강한 O2-, H2O2, OH., nitric oxide (NO), eicosanoids 및 inflammatory cytokines을 비롯한 여러 물질을 분비함으로서 염증 반응을 촉진시켜 숙주방어기전에 중요한 역할을 하고 있으며, 대식세포의 활성화에 의해 미생물, 감 염체 및 종양세포의 성장을 억제하는 것으로 보고되었다(Minnicozzi, et al., 2011; Casserly, et al., 2010). 자유-라디칼 중 NO는 대표적으로 대식세포의 inducible nitric oxide synthase (iNOS) 유전자에 의해 합성되며 면역반응, 혈관확장, 신경전달, 혈소판 응집억제, 세포외 matrix생성의 억제 등 다양한 기능을 매개한다(Greten, et al., 2011). 따라서 구아바의 in vitro 염증 억제 효능 평가를 위하여 염증 반응 지표인 NO 생성량을 측정하였다.

마우스 대식세포주인 RAW 264.7 세포에 염증유발원인 lipopolysaccaride (LPS)를 이용하여 실험법을 구축하였다. 구아바 열수 추출물 (GW), 메탄올 추출물(GM), 에칠아세테이트 추출물(GEA)을 농도별로 처리하여 세포독성을 측정한 다음, 세포독성이 없는 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)의 처리농도를 결정하였다(Fig. 5).

구축된 실험법을 통해 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)와 LPS를 동시 처리하여 NO 생성량을 측정한 결과, GW, GM, GEA의 처리농도에 의존적으로 LPS에 의해 증가된 NO 생성량이 감소하였다(Fig. 6).

NO 생성을 매개하는 유전자인 iNOS의 발현을 조사하였다. RAW 264.7 세포에 LPS와 GW(50-200 ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)를 6시간 처리하여 cDNA를 합성하여 유전자 발현 정도를 측정한 결과 GW, GM, GEA의 처리농도에 의존적으로 LPS에 의해 증가된 iNOS의 발현이 감소하였다(Fig. 7).

(2)염증 관련 유전자 COX-2, iNOS의 유전자 및 단백질 발현 변 화 조사

구아바 잎 열수(GW), 메탄올(GM) 및 에틸아세테이트(GEA) 추출물의 in vitro 염증 억제 효능평가를 위하여 염증 반응에 의해 생성되는 prostaglandin E2(PGE2)의 생성량을 측정하였다. RAW 264.7 세포에 LPS와 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)를 24시간 처리하여 PGE2 생성량을 측정한 결과, GW, GM, GEA의 처리농도 의존적으로 LPS에 의해 증가된 PGE2 생성량이 감소하였다(Fig. 8).

PGE2의 생성을 매개하며, 염증 반응을 유발하는 유전자인 cyclooxygenase-2 (COX-2)의 유전자 및 단백질 발현을 조사하였다. RAW 264.7 세포에 LPS와 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)를 2시간 (유전자 발현) 및 16시간 (단백질 level) 처리하여 확인한 결과, GW, GM, GEA의 처리농도 의존적으로 LPS에 의해 증가된 COX-2의 유전자 및 단백질 발현이 감소하 였다(Fig. 9.10).

COX-2 발현에 관여하는 promoter의 활성을 확인하기 위해 COX-2 promoter를 세포에 형질전환시킨 뒤 LPS와 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)를 처리하여 확인한 결과, GW, GM, GEA의 처리농도 의존적으로 LPS에 의해 증가된 COX-2의 promoter 활성이 감소하였다(Fig. 11).

(3)염증반응 관련 사이토카인의 발현 및 생성량 변화 조사

COX-2와 iNOS 외에 염증 반응을 매개하는 사이토카인들의 생성량 및 유전자 발현에 대한 구아바 잎 열수(GW), 메탄올(GM) 및 에틸아세테이트(GEA) 추출물 영향을 조사하였다.

LPS와 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)를 시간별로 처리하여 대표적인 염증성 사이토카인으로 TNF-α, IL-6, IL-1β에 대한 영향을 측정하였다. GW, GM, GEA의 처리농도 의존적으로 LPS에 의해 증가된 TNF-α, IL-6, IL-1β 유전자 발현이 일괄적으로 감소하였다(Fig. 12~14).

이들 염증성 사이토카인의 유전자 발현에 대한 영향을 측정한 결과 GEA에서 특이적으로 더욱 효과가 높은 것을 확인하였다.

GW, GM, GEA의 처리농도 의존적으로 LPS에 의해 증가된 TNF-α, IL-6, IL-1β 단백질 생성량 이 일괄적으로 감소하였다(Fig. 15-17). 이들 염증성 사이토카인 단백질 생성량에 대한 영향을 측 정한 결과 GEA에서 특이적으로 더욱 효과가 높은 것을 확인하였다.

(4)항염증 반응 관련 전사조절인자 조사

구아바 잎 열수추출물(GW), 메탄올추출물(GM)과 에틸아세테이트 분획물(GEA)의 COX-2 발현 조절은 대표적으로 NF-κB 전사조절인자에 의해 영향을 받으므로 이들 추출물의 NF-κB 전사활성에 대한 영향을 조사하였다. NF-κB luciferase vector를 세포에 형질전환 시킨 뒤 LPS와 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)를 처리하여 NF-κB 전사활성을 확인한 결과 GW, GM, GEA의 처리농도 의존적으로 LPS에 의해 증가된 NF-κB 전사활성이 감소하였다(Fig. 18,19).

3)In vitro 알러지 억제활성 조사

 

면역질환은 알러지 유발물질 (allergen)이 비만세포 (mast cell)나 호염기구 (basophil)의 세포막에 존재하는 Fc receptor에 매개되어 세포의 핵에서 phospholipaseA2나 lipoxigenase 등을 방출시키고 연쇄적으로 arachidonic acid를 생산하여 결과적으로 leukotriene의 세포외 방출, lysosome을 통하여 histamine이 외부로 방출되어 2차적인 면역이상 반응을 유도하거나 면역 관련의 세포에 손상을 일으키는 것으로 밝혀졌다(Wesolowski and Paumet, 2011; Mukherjee and Zhang, 2011).

과민성면역질환인 알러지 반응은 IgE 항체로의 class switch를 유도하는 cytokine인 IL-4 및 eosinophil의 증식 및 분화를 촉진시켜 eosinophilia를 초래하는 cytokine인 IL-5가 필수적으로 관여하여 근원적 원인인 IgE 항체를 생성한다(Sin and Togias, 2011.).

 

 

 

(1)비만세포의 탈과립 방출량 및 히스타민 생성량 변화조사

생성된 IgE는 비만세포의 IgE receptor, FcεRI에 항원으로 작용하여 비만세포에서 면역반응을 일어나게 하여 탈과립이 일어나고 histamine이나 leukotrien 등의 protein을 분비되므로 탈과립 방출량과 분비된 histamine의 양을 조사하였다. 비만세포주인 RBL-2H3 세포에 알러지 반응의 지표인 탈과립 (degranulation) 방출량과 히스타민 생성량을 측정하기 위하여 DNP-IgE로 세포를 감작시키고 알러젠인 DNP-BSA 처리하는 실험법을 구축하였다. 구아바 열수 추출물(GW), 메탄올 추출물(GM), 에틸아세테이트 분획물(GEA)을 RBL-2H3 세포에 농도별로 처리하여 세포독성을 측정한 뒤 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml)의 처리농도를 결정하였다(Fig. 20).

구축된 실험법을 이용하여 비만세포에서의 탈과립 방출량 및 히스타민 생성량 변화를 조사한 결과, GW, GM, GEA의 처리농도에 의존적으로 DNP-BSA에 의해 증가된 탈과립 방출량(Fig. 21)과 히스타민 생성량 (Fig. 22)이 감소하였다.

(2)과민성 면역반응 관련 사이토카인의 발현 및 생성능 조사

알러지성 질환은 IgE에 의해 매개되고 Type 2 T-helper (Th2) cell, 비만세포 및 eosinophil가 알러지 유발 과정에 중요한 역할을 하며 알러젠에 의해 활성화된 Stat6-매개신호경로를 통한 IL-4의 생성을 통해 Th2 세포들은 IL-4 및 IL-5 등의 알러지와 관련된 cytokine을 분비하게 된다.

IL-4 및 IL-5는 B세포에 의한 IgE 및 IgG의 생성을 유도하고, 골수에서의 산성백혈구의 발달을 자극하여, 염증이 생긴 조직으로 모이도록 유도하고 Th2 세포가 관여하는 Th2 면역반응에 대한 과민성 면역질환의 유도에 핵심적인 역할을 한다.

구아바 잎 열수추출물(GW), 메탄올추출물(GM)과 에틸아세테이트 분획물(GEA)의 알러지 반응을 매개하는 cytokine인 IL-4 및 eosinophil의 증식 및 분화를 촉진시켜 eosinophilia를 초래하는 cytokine인 IL-5에 대한 영향을 조사하였다. 비만세포에 DNP-IgE로 세포를 감작시키고 GW(50-200ug/ml), GM(10-50ug/ml), GEA (10-50ug/ml)와 알러젠인 DNP-BSA를 처리하여 IL-4의 유전자 발현, 단백질 생성량 및 promoter 활성에 대한 영향을 측정하였다. DNP-BSA에 의해 증가된 IL-4의 유전자 발현, 단백질 생성량 및 promoter 활성이 GW(50-200ug/ml), GM(10-50ug/ml), GEA(10-50ug/ml) 농도 의존적으로 감소하였으며, GEA의 효과가 가장 강하였다(Fig. 23~25).

6. 구아바 잎의 in vivo 아토피 피부염 억제활성

 

현재까지 국내․외적으로 인체 아토피성 피부염과 동일한 질환 모델 동물은 없으며, 가장 유사한 모델로 많은 연구자들이 아토피성 피부염 연구에 NC/Nga mouse를 가장 보편적으로 사용하고 있다.

NC/Nga mouse는 자발적 아토피성 피부염 유발 모델로 잘 알려져 있으며, 그 원인으로는 JAK3 kinase의 과활성으로 인한 IgE의 생성이 항진되어 나타난 결과로 보고 된 바 있으나 현재까지도 정확한 기전은 알려져 있지 않다. 이런 기전상의 불분명성에도 불구하고 인간의 아토피성 피부염을 가장 잘 반영하는 실험모델로 약리실험에 널리 이용되고 있다(Matsumoto, et al., 1999). 일본에서는 hapten 형성 물질인 1-chloro 2,4,6-trinitro benzene (picryl chloride)을 이용하여 피부염을 인위적으로 유도하여 이러한 문제를 해결하고 있으나, picryl chloride의 경우 국내 반입이 금지되어 있으므로, 구조적으로 picryl chloride와 유사하여 같은 기전으로 작용하는 1-chloro 2,4-dinitrobenzene (DNCB)을 이용하여 시험을 진행하였음. DNCB를 이용하여 아토피성 피부염의 주된 증상들 (가려움, IgE증가, 피부염증)을 관찰할 수 있고, 아토피성 피부염의 치료제에 의해 증상이 감소되는 현상을 관찰할 수 있음 (Im, et al., 2011; Kim, et al., 2011).

따라서 본 연구에서는 건조 구아바 잎을 100℃ 24시간 열수추출하여 농축 및 동결건조한 후 in vivo 실험을 행하였다

 

.

 

 

1)아토피 동물모델인 NC/Nga 마우스에서의 홍반, 출 혈, 건조, 찰상, 짓무름등의 dermatitis scoring 결과 수집

 

아토피 피부염 동물모델에서 구아바 추출물에 의한 아토피 피부염 억제 효과를 측정하기 위하여 마우스에서 표피 두께, 진피 두께, 염증성 세포의 침윤 등의결과를 측정하였다.

아토피 피부염의 임상적 특징은 크게 소양증 (국한성 또는 전신성 가려움), 발진, 만성적 재발 등을 들 수 있는데 혈청내 IgE 수치 향상, eosinophilia, basophilia의 자발적인 히스타민 분비 증가, CD23 발현 증가, CD8 suppressor 감소, Th2에 의한 IL-4, IL-5 분비 증가 등의 현상을 동반하기도 한다. DNCB에 의한 아토피 피부염 유발에 의해 NC/Nga 마우스의 체중이 감소하였으며, 구아바 추출물 처리군의 마우스에서는 체중이 회복되었다(Fig. 28). 또한 DNCB에 의해 증가된 귀의 두께는 구아바 추출물 처리에 의해 감소됨을 확인하였다(Fig. 29).

2)마우스 혈청에서의 과민성 면역반응 관련 사이토카 인의 발현 및 생성능조사

 

아토피 피부염 유발로 증가된 IgE양을 NC/Nga 마우스의 혈청을 분리하여 측정한 결과, DNCB에 의해 증가된 IgE 양이 구아바 추출물에 의해 유의적인 감소 효과를 나타내었다(Fig. 30).

또한 아토피 피부염 유발에 의해 변화되는 IL-10 생성량을 NC/Nga 마우스 혈청에서 확인한 결과, DNCB에 의해 감소된 IL-10 생성량이 구아바 추출물에 의해 유의적인 증가 효과를 나타내었다(Fig. 31, 32).

그리고 아토피 피부염과 같은 면역반응의 유발에 의해 IL-4와 TNF-α 유전자 발현에 변화를 확인하기 위해 마우스 귀 조직에서 cDNA를 얻어 이를 확인하였다. DNCB 투여에 의한 IL-4와 TNF-α 유전자 발현이 구아바 추출물에 의해 감소하였다(Fig. 33).

3)Hematoxylin & eosin 염색 후 조직병리학적 방법 을 이용하여 아토피 피부염 억제 여부 측정

 

아토피 피부염이 유발되었을 때 마우스 피부 조직의 변화를 관찰하기 위하여 마우스 귀를 hematoxylin과 eosin으로 염색하여 확인한 결과 DNCB에 의해 증가된 피부 조직 (위부터 각질, 표피, 진피, 연골 층)과 진피층에서 염증세포의 침윤이 구아바 추출물의 농도 의존적으로 피부조직의 두께와 진피층의 염증세포의 침윤이 감소하였을 관찰할 수 있었다(Fig.34).

7. 구아바 잎 발효액의 아토피 피부염 억제활성

 

구아바 생잎을 세척하여 물기를 제거한 후 설탕을 1:1비율로 첨가·혼합한 것을 실온에서 1시간 숙성시킨 다음에 여과하여 구아바 잎 발효액을 제조하였다. 이 구아바 잎 발효액은 30Brix, pH3.94, 산도 0.24의 물성을 가지고 있고, 또한 총 폴리페놀이 8.81mg%, flavonoid가 1.47mg% 함유되어 있으며, 특히 예비실험에서 항산화 활성도 우수한 것으로 밝혀졌다.

따라서 구아바 잎 발효액의 에탄올 분획을 사용하여 in vitroin vivo 아토피 피부염 억제활성을 조사하였다.

 

1)구아바 잎 발효액의 in vitro 아토피 피부염 억제활 성 조사

 

피부에서의 염증반응 과정은 Th2 chemokine들에 의존적으로 일어나며, chemokine 들은 면역세포들을 조절하는데 관여함. TARC는 Th2 유형의 chemokine으로 피부세포, 수지상세포, 내피세포, 상피세포 등에서 만들어진다(Homey, et al., 2007).

Macrophage-derived chemokine (MDC/CCL22)은 TARC와 밀접하게 관련되어 있으며, 수지상세포, B 림프구, 대식세포, 피부세포, 내피세포 등에 의해 생성된다. 최근의 연구에 따르면 아토피 피부염 환자의 혈청내 TARC와 MDC chemokine의 양에 따라 아토피 피부염 유발이 결정된다고 알려져 있으며, 이들 chemokine들에 의해 여러 가지 질병들이 조절된다. Cutaneous T cell-attracting chemokine (CTACK/CCL27)는 피부에서 선택적으로 발현되며 아토피 피부염을 조절하는 chemokine이다 (Yamashita and Kuroda, 2002).

본 실험에서는 구아바 잎 발효 에탄올 분획에 대한 아토피 피부염의 억제 효능을 평가하기 위해 먼저 구아바 에탄올 분획 (G-EtOH), 구아바 발효 에탄올 대조군 분획 (GF-C-EtOH) 및 구아바 발효 에탄올 분획 (GF-EtOH)에 대한 세포독성을 확인하였다. 구아바 잎 에탄올 분획의 경우 10㎍/ml, 구아바 발효 에탄올 대조군 분획과 구아바 발효 에탄올 분획의 경우 100㎍/ml 까지 세포독성이 없는 것을 확인하였다(Fig. 35).

구아바 발효 분획이 인체 피부 각질 세포에서 아토피 피부염 관련 유전자 (TARC, MDC, CTACK)의 발현에 미치는 영향을 조사하였다. TNF-α와 IFN-γ에 의해 증가된 TARC, MDC 및 CTACK의 유전자 발현이 구아바 에탄올 분획과 구아바 발효 에탄올 분획의 처리 농도 의존적으로 감소하였다. 구아바 에탄올 분획의 경우 TNF-α와 IFN-γ에 의해 증가된 TARC, MDC 및 CTACK의 유전자 발현에 영향이 없었다(Fig. 36).

HaCaT 세포에 pGL3-NF-κB-Luc vector를 형질전환 시킨 뒤 구아바 잎 발효 에탄올 분획 (GF-EtOH)과 TNF-α와 IFN-γ을 24시간 처리한 뒤 NF-κB luciferase activity에 대한 영향을 조사한 결과, TNF-α와 IFN-γ에 의해 증가된 NF-κB 전사활성이 구아바 잎 발효 에탄올 분획의 농도 의존적으로 감소하였다(Fig. 37).

핵 내로 이동되는 NF-κB 단백질 양을 측정한 결과 TNF-α와 IFN-γ에 의해 증가된 NF-κB p65의 핵 내로의 translocation이 구아바 잎 발효 에탄올 분획 (GF-EtOH)의 농도 의존적으로 감소하였다 (Fig. 38). 또한, IκB 단백질의 양의 경우 TNF-α와 IFN-γ에 의해 degradation 되어 감소하며 구아바 잎 발효 에탄올 분획 (GF-EtOH)에 의해 회복됨을 확인하였다(Fig. 39).

Signal transducers and activators of transcription 1 (STAT1)은 IFN-γ와 같은 사이토카인의 신호전달에 의해 활성화되어 각질세포에서 면역반응을 조절하는 핵심 전사조절인자이다. 구아바 잎 발효 에탄올 분획 (GF-EtOH)의 TARC의 생성 및 유전자 발현 억제효능 조절기전을 조사하기 위하여 STAT1에 대한 영향을 조사하였다.

TNF-α와 IFN-γ에 의해 증가된 STAT1의 Tyr 701 잔기의 인산화가 구아바 잎 발효 에탄올 분획 (GF-EtOH)의 농도 의존적으로 감소하였다(Fig. 41).

구아바 잎 발효 에탄올 분획의 전사조절인자 STAT1 조절 여부를 확인하기 위해 STAT1 inhibitor인 AG490을 처리하여 아토피 피부염 지표인 TARC 유전자 발현에 대한 영향을 측정하였다. TNF-α와 IFN-γ에 의해 증가된 TARC 유전자 발현이 구아바 잎 발효 에탄올 분획 (GF-EtOH)과 AG490의 단독과 동시처리에 의해 감소하였다(Fig. 42).

2)구아바 잎 발효액의 in vivo 아토피 피부염 억제성

조사

 

일본에서는 hapten 형성 물질인 1-chloro-2,4,6-trinitro benzene (picryl chloride)을 이용하여 피부염을 인위적으로 유도하여 이러한 문제를 해결하고 있으나, picryl chloride의 경우 국내 반입이 금지되어 있으므로, 구조적으로 picryl chloride와 유사하여 같은 기전으로 작용하는 1-chloro 2,4-dinitroben

zene (DNCB)을 이용하여 시험을 진행하였다. DNCB를 이용하여 아토피성 피부염의 주된 증상들 (가려움, IgE증가, 피부염증)을 관찰할 수 있고, 아토피성 피부염의 치료제에 의해 증상이 감소되는 현상을 관찰할 수 있다.

아토피피부염 유도 및 구아바 발효 추출물 처치 방법은 다음과 같이 행하였다.

6주령 NC/Nga 마우스를 중앙실험동물로부터 구입하여 2주간 안정화 단계를 거친 후 실험하였으며, 1% 또는 0.2%의 DNCB는 등과 귀에 도포하여 아토피 피부염을 유발하였다. 그 후 구아바 잎 발효 시료에 대한 억제 효능을 알아보기 위해 구아바 잎 발효 시료를 농도별로 등과 귀에 도포하였으며 실험모식도는 아래와 같다(Fig. 43).

(1)아토피 동물 모델인 NC/Nga 마우스에서의 홍반, 출혈, 건조 등 의 dermatitis scoring 결과 수집

먼저 아토피 피부염 동물모델에서 구아바 발효 시료에 의한 아토피 피부염 억제 효과를 조사하기 위해 마우스에서의표피 두께, 진피 두께, 염증성 세포의 침윤 등의결과를 측정하였다.

DNCB에 의한 아토피 피부염 유발에 의해 NC/Nga 마우스의 체중이 감소하였으며, 구아바 잎 발효 시료 처리에 의해 마우스의 체중이 회복하였다(Fig. 44). 또한 DNCB에 의해 증가된 귀의 두께는 구아바 발효 시료 처리에 의해 감소하였다(Fig. 45).

(2)마우스 혈청에서의 과민성 면역반응 관련 사이토카인의 발현

및 생성량변화 조사

아토피 피부염 유발로 증가된 IgE양을 NC/Nga 마우스의 혈청을 분리하여 측정한 결과, DNCB에 의해 증가된 IgE 양이 구아바 잎 발효 시료에 의해 농도 의존적으로 감소하였다(Fig. 46).

아토피 피부염 유발에 의해 변화되는 IL-10 생성량을 NC/Nga 마우스 혈청에서 확인한 결과, DNCB에 의해 감소된 IL-10 생성량에 대해 구아바 잎 발효 시료는 영향이 없었다(Fig. 47).

아토피 피부염 유발로 증가된 IgE 생성량을 마우스의 혈청에서 측정한 결과, DNCB에 의해 증가된 IgE 생성량이 구아바 잎 발효 시료 농도 의존적으로 감소하는 것을 확인할 수 있었다(Fig. 48).

아토피 피부염과 같은 면역반응의 유발에 의해 IL-4와 TNF-α 유전자 발현에 변화를 확인하기 위해 마우스 귀 조직에서 cDNA를 얻어 이를 확인하였다. DNCB 투여에 의한 IL-4와 TNF-α 유전자 발현이 구아바 잎 발효 시료에 의해 감소하였다(Fig. 49).

8. 구아바 잎 발효액을 이용한 항아토피 식품 개발

 

1)구아바 잎 발효액의 대량생산

 

아토피 피부염 억제 활성이 규명된 구아바 잎 발효액의 대량생산 공정은 다음 Fig. 50과 같다. 먼저 구아바 잎의 전처리(세척, 절단)하여 95~100℃에서 8시간 열수추출한 후, 농축하여 10Brix로 조절한 후 sucrose 5%와 효소추출물 0.5%를 가한 다음 살균하여 Saccharomyces cerevisiaeLactobacillus cerevisiae 또는 Bacillus subtilis를 접종하였다. 이것을 30~40℃에서 3~5일간 배양한 후 발효액을 조제하였다.

구아바 잎 열수추출물에 Lactobacillus plantarum을 접종하여 생물전환된 것을 살균·여과·농축·동결건조한 시료에서 항아토피 활성이 비교적 높게 나타났다. 이 발효대사체를 이용하여 구아바를 주원료로하는 가공식품 개발을 위한 구아바 주요원료의 배합비율은 Table 5와 같다.

(1)구아바 과립차 제조를 위한 당 발효액 개발

구아바 잎과 열매의 polyphenol성 물질, flavonoid, carotenoid등 다양한

생리활성물질, 그리고 비타민, 무기질, 식이섬유 등 풍부한 영양물질을 이

용한 구아바 과립차를 개발하기 위하여 구아바 잎, 열매, 가지를 이용하

여 당 발효액을 다음과 같이 제조하였다(Table 6).

(2)구아바 당 발효액의 특성

구아바 과립차 제조에 사용될 구아바 당 발효액의 특성은 Table 6과 같

이 제조된 구아바 열매 당 발효액의 고형물 함량은 26.8Brix, pH 3.94, 구

아바 잎 당 발효액은 25.3Brix, pH 4.38, 구아바 가지 당 발효액은

41.0Brix, pH 3.97, 구아바 과일 flavor는 53.9Brix, pH 3.12로 나타났다

(Table 7).

구아바 열매와 잎의 고형물 함량이 가지보다 낮은 이유는 자체 수분함량

이 높은 것에 기인되는 것으로 사료된다. 잎의 pH가 높은 이유는 잎에

다량 함유된 tannin 등 폴리페놀성 물질에 의해 미생물 생육이 억제되어

산 생성이 저해된 것으로 추측된다.

(3)구아바 당 발효액의 구성비율

먼저 구아바 열매, 잎, 가지 당 발효액의 구성 비율을 Table 8과 같이 5

: 3 : 2로 혼합하여 물성을 조사하였다. 혼합 당 발효액의 Brix는 29.6,

pH 3.9로서 과립차 제조를 위한 적정 물성인 것으로 판단된다. 그리고 고

형물 함량을 높이기 위하여 보당 또는 부형재 첨가가 요구된다.

(4)동결건조 구아바 열매분말 첨가제 따른 혼합발효액의 특성

구아바 당 혼합 발효액은 과립차 제조에 필요한 고형물 함량이 다소 낮

아서 동결건조 혼합 구아바 열매 분말을 5%, 10%, 15%, 20%씩 혼합 용

해하여 물성 등을 조사하였다(Table 9).

동결건조 구아바 열매 분말의 첨가농도가 증가 할수록 고형물 함량은 점

진적으로 증가하였고, pH는 약간 증가 하였으며, 기호성은 향상되는 특징

을 보였다. 동결건조 분말 15~20% 이상 첨가하여야 과립차 제조 시 향

미, 기호성, 영양성 등에 있어서 양호한 결과가 도출될 것으로 예상된다.

(5)구아바 열매 flavor 첨가량에 대한 기호성 비교

구아바 당 혼합 발효액에 구아바 열매 flavor을 0.5%, 1%, 1.5%, 2%, 3

%, 4% 씩 첨가하여 향미 검사를 행한 결과 3%이상 첨가 시에 과립차

제조에 적합한 향미가 나타났다(Table 10).

따라서 과립차 제조 시에 3~4%이상의 flavor를 첨가하여 시제품을 제조

Ⅳ. 요 약

본 연구는 구아바 잎과 열매의 이화학적 특성을 조사한 후 구아바 잎 유래 열수추출물, 메탄올 추출물, 에틸아세테이트 분획물의 in vito 및 in vivo 항아토피 활성을 조사하였고, 그리고 최종적으로는 구아바 잎 발효액의 아토피 피부염 억제활성을 검증하였다. 건조 구아바 잎은 수분 7.01%, 탄수화물 15.65%, 조단백질 5.48%, 조지방 2.16%, 회분 9.12%, 식이섬유 30.28%로 구성되어 있었고, 그리고 구아바 열매에는 수분 80.18%, 탄수화물 17.41%, 조단백질 1.23%, 조지방 0.40%, 회분 0.78%, 식이섬유 3.45%로 나타났다. 건조 구아바 잎의 무기질은 Ca이 1925.62mg%로 가장 높았고 K>Na>Mg>P>Fe순으로 높게 검출된 반면에 열매에는 K가 314.62mg%로 가장 높았고 Na>P>Ca>Mg>Fe순으로 검출되었다.

건조 구아바 잎의 비타민은 niacin이 32.55mg%, 비타민 C가 13.27mg%, 비타민 B1, B2, B6가 각각 0.03, 0.03, 0.05mg% 함유되어 있었고, 그리고 열매에는 niacin 41.17mg%, 비타민C 45.28mg%, 비타민 B1 0.01mg%, B2 0.08mg%, B6 0.09mg%로서 잎보다 비타민 함량이 전반적으로 높게 나타났다. 건조 구아바 잎은 항산화물질인 총 폴리페놀과 카로틴노이드 함량이 각각 3.15mg, 미량으로서 열매의 총 폴리페놀 490.13mg%, 카로틴노이드 215.28mg%에 비해 낮게 검출되었다.

구아바 잎 유래 유용 추출물의 항아토피 활성 검증을 위해 먼저 구아바 잎으로부터 추출한 열수추출물(GW), 메탄올 추출물(GM), 그리고 에탈아세테이트 분획물(GEA)을 건조하여 in vitro 아토피 피부염의 억제활성, 즉 TARC/CCL17의 발현능, in vitro 염증 억제활성, 그리고 in vitro 알러지 억제활성을 조사하였다. 인체 피부 각질세포인 HaCaTtpvh에 TNF-α를 처리하여 아토피 지표인 TARC/CCL17을 유도시킨 후, 구아바 잎 열수추출물(GW), 메탄올 추출물(GM), 및 에탈아세테이트 분획물(GEA)를 10,50,100,200μg/mL씩 처리하여 TARC/CCL17의 생성능을 측정하였다.

TNF-α에 의해 증가된 TARC/CCL17의 유전자 발현량과 단백질 생성량이 GW, GM, GEA의 처리 농도에 의존적으로 감소하였다. 또한 TNF-α에 의해 증가된 RANTES/CCL5의 생성량이 GW, GM, GEA 처리에 따라 농도 의존적으로 감소하였다. 항아토피 반응 관련 전사조절인자 NF-κB 및 상위신호전달 체계인 MAP kinases의 활성이 감소하였다.

 

구아바 잎 추출물(GW, GM, GEA)의 in vitro 염증 억제활성을 평가하기 위하여 염증 지표인 No 생성량을 먼저 측정하였다. 구축된 실험법을 통해 GW(50~200μg/mL), GM(10~50μg/mL), GEA(10~50μg/mL)와 LPS를 동시에 처리하여 No 생성능을 측정한 결과 GW, GM, GEA의 처리농도에 의존적으로 LPS에 의해 증가된 No 생성량이 감소하였다.

그리고 No 생성을 매개하는 유전자인 iNOS의 발현도 GW, GM, GEA의 처리농도에 의존적으로 감소하여, 구아바 잎 추출물의 항염증 활성을 확인하였다. 또한 염증 반응에 관여하는 prostaglandin E2(PGE2)의 생성량을 측정하기 위해 RAW 264.7 세포에 LPS와 구아파 잎 추출물 GW(50~200μg/mL), GM(10~50μg/mL), GEA(10~50μg/mL)를 24시간 처리하였을 때 GW, GM, GEA의 처리농도 의존적으로 PGE2 생성량이 감소하였다. PGE2 생성을 매개하며 염증반응을 유발하는 유전자인 cyclocoxygenase-2(COX-2) 유전자의 발현과 단백질 생성량 및 COX-2 promoter 활성도 구아바 잎 추출물의 처리로 감소하였다.

그리고 COX-2 및 iNOS 외에 염증반응을 매개하는 cyrokine 생성량과 유전자 발현에 대한 구아바 잎 추출물(GW, GM, GEA)의 영향을 조사하였다. 구아바 잎 추출물의 처리시에 대표적인 염증성 cytokine인 TNF-α, IL-6, IL-1β의 유전자 발현과 단백질 생성량이 높게 나타났다. COX-2 발현을 조절하는 전사조절인자인 NF-κB3의 활성도 억제되었음을 확인하였다.

 

비만 세포주인 RBL-2H3 세포에 알러지 반응의 지표인 탈과립(degranulation) 방출량과 히스타민 생성량을 조사하기 위하여 DNP-IgE 세포를 감작시키고 알러젠인 DNP-BSA로 처리하는 실험법을 구축한 다음, 구아바 잎 추출물(GW, GM, GEA)을 처리하였을 때 처리농도에 의존적으로 탈과립 방출량과 히스타민 생성량이 감소하였다. 또한 과민성 알러지 반응을 매개하는 cytokine IL-4 및 IL-5의 유전자 발현량, 단백질 생성량 및 promoter 활성들이 모두 농도의존적으로 감소함을 확인하였다. 또한 알러지 반응 관련 전사조절인자(IL-4 및 IL-5)의 활성도 억제되었다.

 

자발적 아토피성 피부염 유발 동물모델인 NC/Nga mouce를 이용하여 in vivo 아토피성 주된 증상인 가려움, IgE증가, 각종 피부염증 등을 건조 구아바 잎 열수추출물(100℃, 24시간 추출, 농축, 동결건조)을 100mg/mL 및 200mg/mL 처리에 따른 효과를 조사하였다.

DNCB(1-chloro-2.4-dinitrobenzene)로 유발된 아토피 피부염 동물 모델인 NC/Nga 마우스의 체중이 감소하였으나 구아바 잎 열수추출물 처리로 체중이 회복되었다. 또한 DNCB에 의해 증가된 귀의 두께는 구아바 추출물 처리로 감소되었음을 확인하였다. NC/Nga 마우스의 혈청을 분리하여 IgE과 IL-10생성량을 측정한 결과, 구아바 잎 열수추출 처리로 DNCB에 의해 증가된 IgE양이 감소하였고, IL-10 생성량은 유의적인 증가 효과를 보였다.

그리고 아토피 피부염과 같은 면역 반응의 유발에 의해 증가되는 IL-4와 TNF-α 유전자의 발현은 DNCB 투여로 wd가된 것이 구아바 잎 열수추출물에 의해 감소되었다. 아토피 피부염의 유발로 발생하는 NC/Nga 마우스의 피부조직을 관찰하기 위해 마우스의 귀를 hematoxylin과 eosin으로 염색하여 확인한 결과, DNCB에 의해 증가된 피부조직(각질, 표피, 진피, 연골층)에서 염증세포의 침윤이 구아바 잎 열수 추출물의 농도 의존적으로 감소되었다.

 

구아바 잎 발효액 제품(총 폴리페놀 8.81mg%, 폴라보노이드 1.47mg%)의 에탄올 분획을 사용하여 아토피 피부염의 in vitro 및 in vivo 억제활성을 조사하였다.

피부 각질 세포인 HaCaT세포에 구아바 발효액 에탄올 분획물을 처리한 결과 nitric oxide 생성량이 감소하였다. 염증 관련 COX-2와 iNOS의 발현 및 염증반응 관련 사이토카인의 발현 및 생성량도 억제되었다. 항염증 반응 관련 전사조절인자의 활성이 억제되엇음을 확인하였다.

비만세포에 구아바 발효액 에탄올 분획물을 처리한 결과 알러지 유발 인자인 histamine의 생성 및 알러지 반응에 의해 일어나는 degranulation 양이 감소하였다. 과민성 면역 반응 관련 사이토카인의 발현 및 생성량이 감소하였으며, 항알러지 반응 관련 전자조절인자의 활성도 억제되었다. 아토피 동물모델에 구아바 발효액 에탄올 분획물을 처리한 결과 아토피 피부염 유발로 증가된 홍반, 출혈, 건조 등의 dermatitis scoring가 감소하였다. 과민성 면역반응 관련 사이토카인의 발현 및 생성량이 감소하였고, 조직병리학적인 소견에서 피부조직의 두께와 염증세포의 침윤이 억제되었음을 관찰할 수 있었다.

그리고 상기 구아바 잎 발효액과 구아바 열매 동결건조 분말, 구아바 당침액 등을 주원료로 하는 항아토피 과립차를 개발하였다.

Study on the Antiatopic Dermatitis of Psidium guajave Leaf

This study was carried out to investigate the physicochemical characteristics and antiatopic dermatitis of Psidium guajava extracts

Proximate compossition, minerals Total polyphenol, carotenoide and vitamin contents of Psidium guajava leaf dried powder, and sugar fruit were examined.

The Psidium guajava leaf consits of 7.01% of moisture, 15.65% of carbohydrate, 5.48% of crude protein, 2.16% of crude lipid, 9.12% of ash and 30.28% of diatery fiber. Mineral contents of Psidium guajava leaf were in the order of Ca>K>Na>Mg>P>Fe.

The niacin content was the longest as 32.55mg% for Psidium guajava leaf vitamins followed by vitamin C(13.27mg%), vitamin B6(0.05mg%), vitamin B1(0.03mg%), vitamin B2(0.03mg%)

The total polyphenol and carotenoid contents of Psidium guajava leaf were 3.15mg% and trace, respectively.

The moisture, carbolydrate, crude protein, crude lipid, ash and dietary fiber contents of Psidium guajava fruit were 80.18, 17.41, 1.23, 040, 0.78 and 3.45%, respectively.

The K content was the longest for Psidium guajava fruit minerals followed by Na, P, Ca, Mg, Fe.

The vitamin C content was the highest as 45.28mg% in Psidium guajava fruit follwed by niacin, vit B6, B2 and B1.

The total polyphenol and carotenoid contents of Psidium guajava fruit were 490.13mg% and 215.28mg%, respectively, and carotenoid was not detected in Psidium guajava leaf.

The inhibitory effects of Psidium guajava leaf extracts such as hot water extract, methanol extract and ethylacetate extract in the development of atopic dermatitis were also assayed

Psidium guajava (P. guajava) is an important food crop and medicinal plant with antioxidant, antiinflammatory, and anti-allergic activities, supporting its traditional uses. However, its precise effects remain unknown. We investigated the effects of P. guajava ethyl acetate extract (PGEA) on IgE-mediated allergic responses in rat mast RBL-2H3 cells.

PGEA reduced antigen (DNP–.BSA)-induced release of bhexosaminidase and histamine in IgE-sensitized RBL-2H3 cells. In addition, it inhibited antigen-induced IL-4 and TNF-a mRNA expression and protein production in IgE-sensitized RBL-2H3 cells. PGEA also suppressed antigen-induced COX-2 mRNA and protein expression in these cells, as well as antigen-induced activation of NFAT and reactive oxygen species. Moreover, it inhibited antigen-induced activation of NFjB and degradation of IjB-a. To identify the mechanisms underpinning the inhibition of degranulation and cytokine production by PGEA, we examined the activation of intracellular FceRI signaling molecules.

PGEA suppressed antigen-induced phosphorylation of Syk, LAT, Gab2, and PLCc2 but not Lyn, and inhibited antigen-induced phosphorylation of downstream signaling intermediates including MAP kinases and Akt. Collectively, the anti-allergic effects of PGEA in vitro suggest its possible therapeutic application to inflammatory allergic diseases, in which its inhibition of inflammatory cytokine production and FceRI-dependent signaling events in mast cells may be hugely beneficial.

Psidium guajava (P. guajava) is a food and medicinal plant with antioxidant, antiinflammatory, and anti-allergic activities that support its traditional uses. The aim of this study was to determine the effects of P. guajava ethyl acetate extract (PGEA) on atopic dermatitis and to investigate the possible mechanisms by which PGEA inhibits cytokineinduced Th2 chemokine expression in HaCaT human keratinocyte cells.We found that PGEA suppressed the IFN-/TNF--co-induced production of thymus and activation-regulated chemokine (TARC) protein and mRNA in HaCaT cells.

Additionally, PGEA inhibited the TNF-/IFN--co-induced activation of NF-B and STAT1 and increased the expression of heme oxygenase-1 (HO-1) protein and mRNA. HO-1 inhibitor enhanced the suppressive effects of PGEA on TNF-/IFN--co-induced TARC production and gene expression. Collectively, these data demonstrate that PGEA inhibits chemokine expression in keratinocytes by inducing HO-1 expression and it suggests a possible therapeutic application in atopic dermatitis and other inflammatory skin diseases.

Atopic dermatitis (AD) is a chronic, relapsing, and inflammatory skin disease associated with eczematous symptoms and IgE hyperproduction. Psidium guajava is an important food crop and medicinal plant with anti-oxidant, anti-inflammatory, and anti-allergic activities, supporting its traditional uses. Our previous studies have shown that P. guajava extract inhibits Th2 chemokine expression by suppressing the activation of NF-jB and STAT1 co-stimulated with TNF-a and INF-c. In this study, we investigated the inhibitory effect of P. guajava water extract(PGW) on 2,4-dinitrochlorobenzene (DNCB)-induced AD-like skin lesions in NC/Nga mice. Treatment of cream containing PGW onto DNCB-induced AD-like skin lesions in NC/Nga mice ameliorated lesion intensity scores, levels of IgE, thymus and activation-regulated chemokine (TARC), TNF-a, and IL-4 in serum and ears. In contrast, PGW increased level of the immunosuppressive cytokine IL-10. Histological analyses demonstrated decreased thickening of the epidermis/

dermis as well as dermal infiltration by inflammatory cells. These results suggest that cream containing PGW may be a potential therapeutic modality for AD and adjunctive agent to control pruritus in AD. Administration of ethanol fraction from Psidium guajava leaf fermentative metabolite onto DNCB-induced AD-like skin lesions in NC/Nga mice ameliorated lesion intensity seores, levels of IgE, thymus and activation-regulated chemokine(TARC), TNF-α, and IL-4 in serum and ears.

In contrast, ethanol fraction from Psidium guajava leaf fermentative metabolite increased level of the immunosuppressive cytokine IL-10.

Histopathological examination showed reduced thickness of the epidermis/dermis and dermal infiltration of inflammatory cells in the ears. In conclusion, the in vitro/in vivo anti-allergic and anti-atopic effects of PGEA suggest possible therapeutic applications for this agent in allergic diseases through the inhibition of inflammatory and atopic dermatitis cytokines
